Standard Operating Procedure – SOP (only in portuguese)
To conduct experiments involving protein extraction, quantification, and digestion, it is essential that laboratory procedures are well established and properly implemented by those responsible. Therefore, this Standard Operating Procedure (SOP) aims to standardize the steps of the protocols for protein extraction (Urea/Thiourea and TCA/Acetone), protein concentration determination using the Bradford method, protein precipitation with Methanol/Chloroform, and protein digestion using the FASP method and gel band digestion. Additionally, this SOP includes guidelines for the proper submission of samples for proteomic analysis. The adoption of well-defined protocols ensures experimental reproducibility, safeguards sample integrity, and minimizes potential variations in the results obtained. Accordingly, this document provides a detailed description of the procedures to be followed, including the solutions used, their respective functions, and the methodologies applied. This aims to ensure an organized and safe research environment, contributing to the efficiency of analyses and the reliability of the data generated.
